Synthetic routes toward benzene 1,3,5-tricarboxamide biofunctionalization

Project Description: 

The water soluble benzene 1,3,5-tricarboxamide (BTA) structural motif is a promising candidate for a synthetic extracellular matrix (ECM) because it dynamically assembles into collagen-like fibers in water. Before a synthetic ECM can be realized, however, the BTA unit must be functionalized with biological molecules at an extended distance from the fiber's core to maintain biological utility. This research describes a 3 step synthetic route towards a monoextended arm in 45% yield. Through UV-spectrophotometry it is demonstrated that this extended arm can still be incorporated into the fibers at 20% molar volume. Further, the extended arm was functionalized with an oxyamine for a future oxime ligation reaction with an oxidized s-protein.
