Folding Kinetics of Chymotrypsin Inhibitor 2

Research Project Description: 

Chymotrypsin Inhibitor 2 (CI2), is a protein found in barley that inhibits proteases and is a small, single domain protein. Various CI2 mutants with a single amino acid substitution at the same location on the protein have been made to test how charge interactions or hydrophobic interactions in the transition state, affect the folding rate. For each mutant, folding and unfolding experiments were done to collect time resolved fluorescence data using varying concentrations of denaturant, in order to obtain the rate constants. These observed rate constants were fit to an equation derived from the linear dependence of folding rate on denaturant concentration. From this, the folding rate in the absence of denaturant can be extrapolated

Enhancing Biology Lessons with Interactive Simulations

Curriculum Project Description: 

After six weeks of research in a biochemistry lab, I observed how instrumental computer models were in visualizing and predicting the folding rates of proteins based on the interactions of amino acid side chains. I found the advanced software used in the lab to view and create polypeptides was indispensable to the progress of the research. Data was collected and entered into a computer to efficiently create complex graphs such as Chevron plots.
My project focuses on the incorporation of computer simulations to add motivation and variety to biology lessons. My goal was to find at least one internet activity per unit that fosters a sense of independent exploration for each student. The simulations will have students set up experiments, collect data, create graphs, and complete worksheets that guide them through each site. A Teachers Guide is included with a list of CA State standards covered, length of time for each activity, and suggestions for use. Additional checks for understanding are provided in the form of quizzes, concept maps, and short writing assignments. These activities can be altered to target the needs of students at any level.
